For even neglecting its biological importance, there would still be a lot of interests in nucleic acid as a fundamental molecule for chemists to study and use as an object for chemical building blocks, sensing, polymer design and others.1 Nowadays, nucleic acids have been emerged from various research areas such as DNA computing, nano-size structures using DNA, sensors recognizing mRNA and DNA, and oligonucleotide therapeutics, and others.2 Chemical modification of nucleic acid systems is essential to obtain more effective and stable materials. From nucleoside to oligonucleotide, chemical modification is open to any part of nucleic acid. We focus on the synthesis of new phosphoramidites to incorporate into the oligonucleotides. In this seminar, we will discuss our research efforts into oligonucleotides functionalized with steroid derivatives and cationic amine moieties to enhance their cell permeability.3 We will also describe about our syntheses of modified oligonucleotides containing fluorescent tags for use as molecular probes (molecular beacons) to detect single-nucleotide polymorphisim (SNP) in nucleic acids,4 and to monitor secondary structures of various nucleic acids such as B-Z transition,5 G-quadruplex structures,6 and DNA i-motif structures.
